Indication
Treatment of depressive illness (including endogenous and psychotic depression), nocturnal enuresis in children, neuropathic pain, and migraine prophylaxis.
Ingredients
- Amitriptyline hydrochloride 25 mg
Direction
Depression — Adults: Initially 25 mg two to three times daily or 50–75 mg as a single night-time dose; maintenance 50–150 mg/day. Elderly: 25–50 mg daily. Neuropathic pain — Adults: 10–25 mg at night, titrated upward as tolerated. Dosing should be individualised and supervised by a physician.
Side effects
- Dry mouth
- constipation
- blurred vision (anticholinergic effects).
- Sedation and drowsiness.
- Postural hypotension.
- Weight gain.
- Tachycardia.
- Urinary retention.
- Confusion
- especially in the elderly.
Precautions
- Do not use with MAOIs — allow at least 14 days after stopping MAOIs.
- Contraindicated in recent myocardial infarction
- arrhythmias
- and severe hepatic impairment.
- Risk of QT prolongation and cardiac arrhythmias.
- May lower seizure threshold — use with caution in epilepsy.
- Avoid abrupt discontinuation — taper gradually.
- Risk of suicidality at treatment initiation
- especially in young adults.
- Avoid alcohol
- may potentiate CNS depression.
- Keep out of reach of children.
