Laroxyl® 25 mg

Indication

Treatment of depressive illness (including endogenous and psychotic depression), nocturnal enuresis in children, neuropathic pain, and migraine prophylaxis.

Ingredients

• Amitriptyline hydrochloride 25 mg

Direction

Depression — Adults: Initially 25 mg two to three times daily or 50–75 mg as a single night-time dose; maintenance 50–150 mg/day. Elderly: 25–50 mg daily. Neuropathic pain — Adults: 10–25 mg at night, titrated upward as tolerated. Dosing should be individualised and supervised by a physician.

Side effects

• Dry mouth
• constipation
• blurred vision (anticholinergic effects).
• Sedation and drowsiness.
• Postural hypotension.
• Weight gain.
• Tachycardia.
• Urinary retention.
• Confusion
• especially in the elderly.

Precautions

• Do not use with MAOIs — allow at least 14 days after stopping MAOIs.
• Contraindicated in recent myocardial infarction
• arrhythmias
• and severe hepatic impairment.
• Risk of QT prolongation and cardiac arrhythmias.
• May lower seizure threshold — use with caution in epilepsy.
• Avoid abrupt discontinuation — taper gradually.
• Risk of suicidality at treatment initiation
• especially in young adults.
• Avoid alcohol
• may potentiate CNS depression.
• Keep out of reach of children.
