Allopurinol 300mg YSP 300mg

Indication

Allopurinol 300 mg (YSP) is a xanthine oxidase inhibitor used for the long-term prevention and management of hyperuricaemia (high uric acid levels) associated with gout, gouty arthritis, uric acid kidney stones, and during chemotherapy of haematological malignancies.

Ingredients

• Allopurinol 300 mg

Direction

Adults: 100–300 mg once daily after meals. Mild gout: 100–200 mg/day. Moderate-to-severe gout: 300–600 mg/day (in divided doses if over 300 mg). Take with plenty of water. Not for use during an acute gout attack.

Side effects

• Skin rash (common — may indicate serious hypersensitivity — discontinue immediately).
• Nausea
• vomiting
• diarrhoea.
• Elevated liver enzymes.
• Stevens
• Johnson syndrome / toxic epidermal necrolysis (rare but life
• threatening).
• Gout exacerbation at start of therapy.

Precautions

• Do not start during an acute gout attack — initiate prophylactic colchicine or NSAID at treatment start.
• Contraindicated in patients with known hypersensitivity to allopurinol.
• Severe skin reactions (Stevens
• Johnson syndrome
• DRESS) — higher risk in patients with HLA
• B*5801 allele (common in Asian populations) — consider testing before initiation.
• Use with caution in renal impairment — dose adjustment required.
• Drug interactions: azathioprine
• mercaptopurine (reduce their dose to 25%)
• warfarin
• theophylline.
• Maintain adequate fluid intake (2–3 L/day).
• Keep out of reach of children.
