Indication
Used to treat major depressive disorder. Also used for neuropathic pain, migraine prophylaxis, and nocturnal enuresis. A tricyclic antidepressant that increases serotonin and norepinephrine activity in the brain.
Ingredients
- Amitriptyline HCl
Direction
Adults (depression): Initially 25–75 mg daily in divided doses or as a single dose at bedtime. Dose may be increased gradually under medical supervision. Elderly: Start at lower doses (10–25 mg at bedtime). Do not stop abruptly.
Side effects
- Very common: dry mouth
- sedation
- drowsiness
- blurred vision
- constipation
- urinary retention.
- Common: dizziness
- postural hypotension
- tachycardia
- tremor
- weight gain.
- Uncommon: cardiac arrhythmias
- confusion (especially in elderly).
- Rare: seizures
- agranulocytosis.
- Very rare: liver toxicity.
Precautions
- Contraindicated in hypersensitivity to amitriptyline or tricyclic antidepressants.
- Contraindicated in recent myocardial infarction
- serious arrhythmias
- and mania.
- Do not use with MAO inhibitors — allow 14 days washout.
- Risk of suicidal thinking in children
- adolescents
- and young adults — monitor closely.
- May prolong QT interval — caution with cardiac disease.
- Causes sedation — impairs driving and operating machinery.
- Anticholinergic effects — caution in urinary retention
- glaucoma
- prostatic hypertrophy.
- Lowers seizure threshold — caution in epilepsy.
- Contraindicated in pregnancy and breastfeeding.
- Keep out of reach of children.
