Indication
Pioglitazone 45 mg is an oral antidiabetic agent (thiazolidinedione/insulin sensitiser) used for the treatment of type 2 diabetes mellitus in adults, as monotherapy (when metformin is inappropriate) or in combination with other antidiabetics including metformin, sulfonylureas, or insulin.
Ingredients
- Pioglitazone
Direction
Adults: The usual starting dose is 15–30 mg once daily, which may be increased to a maximum of 45 mg once daily. Take at the same time each day, with or without food. Dose titration should be guided by a physician.
Side effects
- Fluid retention and oedema.
- Weight gain.
- Hypoglycaemia (especially in combination with insulin or sulfonylureas).
- Headache
- dizziness.
- Increased fracture risk.
- Rare: hepatotoxicity.
Precautions
- Contraindicated in patients with heart failure (NYHA Class I–IV) or history of heart failure.
- Increases risk of fluid retention and oedema
- monitor closely.
- Associated with increased risk of bladder cancer with prolonged use
- do not use in patients with active bladder cancer.
- Associated with increased risk of bone fractures
- particularly in women.
- Not for use in patients with severe hepatic impairment.
- Monitor liver function tests periodically.
- Keep out of reach of children.
