Indication

Domperidone is a dopamine antagonist (prokinetic) used for the symptomatic relief of nausea and vomiting, including nausea and vomiting associated with dopaminergic drugs (e.g., L-dopa). Also used for delayed gastric emptying and gastro-oesophageal reflux.

Ingredients

Direction

Adults: 10 mg up to 3 times daily before meals. Maximum 30 mg per day. Use the lowest effective dose for the shortest duration. Children: dose based on body weight (0.25 mg/kg up to 3 times/day); consult physician.

Side effects

• Dry mouth
• headache.
• Rarely: QT prolongation
• cardiac arrhythmias.
• Rare: extrapyramidal effects (tremor
• tardive dyskinesia)
• galactorrhoea.
• Restlessness
• drowsiness.

Precautions

• Do not use in patients with known hypersensitivity to domperidone.
• Contraindicated in patients with conditions where cardiac conduction is prolonged (QTc prolongation) or concomitant use of QT
• prolonging drugs.
• Avoid in patients with gastro
• intestinal haemorrhage
• mechanical obstruction
• or perforation.
• Use with caution and lowest effective dose in patients over 60 years due to increased cardiac risk.
• Avoid concomitant use of CYP3A4 inhibitors (e.g.
• ketoconazole
• erythromycin).
• Keep out of reach of children.
