Diamide Glibenclamide 5 mg + Metformin HCl 500 mg

Indication

Diamide is a fixed-dose combination tablet containing a sulfonylurea (Glibenclamide) and a biguanide (Metformin HCl) used for the management of type 2 diabetes mellitus in adults when both components are required for adequate blood glucose control.

Ingredients

• Glibenclamide 5 mg
• Metformin hydrochloride 500 mg

Direction

Adults: Dosage individualised by physician based on blood glucose levels. Typically one tablet once or twice daily with meals. Do not exceed the prescribed dose. Take with food to reduce gastrointestinal side effects.

Side effects

• Common: hypoglycaemia (sweating
• tremor
• hunger
• dizziness).
• Common: nausea
• vomiting
• diarrhoea
• abdominal pain (usually transient with metformin).
• Uncommon: skin reactions (rash
• pruritus).
• Rare: lactic acidosis (serious — seek emergency care).
• Rare: blood dyscrasias.
• Very rare: vitamin B12 deficiency with long
• term use.

Precautions

• Contraindicated in type 1 diabetes
• diabetic ketoacidosis.
• Contraindicated in renal impairment (eGFR < 45 mL/min/1.73 m²).
• Contraindicated in hepatic impairment
• adrenal insufficiency.
• Contraindicated in pregnancy and breastfeeding.
• Risk of hypoglycaemia (from glibenclamide component) — caution with missed meals or alcohol.
• Risk of lactic acidosis (from metformin component) — discontinue before contrast procedures.
• Monitor renal function and blood glucose regularly.
• Avoid excessive alcohol.
