Indication
Treatment of depressive illness, nocturnal enuresis in children, neuropathic pain, and migraine prophylaxis.
Ingredients
- Amitriptyline hydrochloride 25 mg
Direction
Depression — Adults: Initially 25 mg two to three times daily or 50–75 mg as a single night-time dose; usual maintenance 50–150 mg/day. Elderly: 10–25 mg/day, adjusted cautiously. Neuropathic pain — 10–25 mg at bedtime, titrated upward. Dosing must be individualised under physician supervision.
Side effects
- Dry mouth
- constipation
- blurred vision (anticholinergic effects).
- Sedation and drowsiness.
- Postural hypotension.
- Weight gain.
- Tachycardia.
- Urinary retention.
- Confusion (especially in the elderly).
Precautions
- Do not use with MAOIs — allow at least 14 days after stopping MAOIs.
- Contraindicated in recent myocardial infarction and arrhythmias.
- Risk of QT prolongation.
- Avoid abrupt discontinuation — taper dose gradually.
- Risk of suicidality
- especially in young adults at treatment initiation.
- Avoid alcohol.
- May impair ability to drive.
- Keep out of reach of children.
