Indication
Colpotrophine (Promestriene 10 mg) is a vaginal oestrogen used for the local treatment of vaginal atrophy and symptoms of urogenital atrophy in peri- and post-menopausal women, including vaginal dryness, itching, burning, and dyspareunia (painful intercourse). Promestriene is a topical oestrogen with very low systemic absorption.

Insert one pessary or apply the cream intravaginally each evening for 20 days, then reduce to 2–3 times weekly for maintenance, or as directed by a physician. Use the applicator provided. Wash hands before and after use.
Side effects
- Local irritation
- burning or itching at application site — uncommon.
- Vaginal discharge — uncommon.
- Systemic oestrogenic effects — very rare due to minimal absorption.
- Allergic reactions — rare.
Precautions
- Do not use if allergic to promestriene or oestrogens.
- Do not use in patients with unexplained vaginal bleeding
- oestrogen
- dependent tumours (breast cancer
- endometrial cancer)
- active or past thromboembolic disease
- or severe liver disease.
- Although systemic absorption is minimal
- periodic gynaecological review is recommended.
- Report any unusual vaginal bleeding to a doctor immediately.
- Consult a doctor before use in patients with history of endometriosis or uterine fibroids.
- Keep out of reach of children.
