Indication
Management of type 2 diabetes mellitus in adults when metformin is inappropriate or insufficient. Pioglitazone improves insulin sensitivity; glimepiride stimulates pancreatic insulin secretion.
Ingredients
- Pioglitazone
- Glimepiride
Direction
Adults: One tablet (30 mg pioglitazone + 4 mg glimepiride) once daily with or immediately before the first main meal of the day. Dose should be individualized based on glycaemic response and tolerability.
Side effects
- Hypoglycaemia (low blood sugar)
- Weight gain
- Fluid retention and oedema
- Anaemia
- Dizziness
- Nausea
- Increased risk of bone fractures
- Upper respiratory tract infection
Precautions
- Contraindicated in type 1 diabetes mellitus or diabetic ketoacidosis.
- Contraindicated in patients with heart failure (NYHA Class I–IV) — pioglitazone may cause fluid retention.
- Contraindicated in patients with active or history of bladder cancer.
- Risk of hypoglycaemia
- especially with missed meals or increased physical activity.
- Monitor liver function periodically.
- Pioglitazone may cause or worsen oedema.
- Associated with increased risk of bone fracture in women.
- Not recommended in severe renal or hepatic impairment.
- Keep out of reach of children.
