Zyloric 100mg

Indication

Zyloric contains Allopurinol, a xanthine oxidase inhibitor used to treat conditions associated with excessive uric acid production or excretion, including gout, uric acid nephrolithiasis, and hyperuricaemia associated with malignancy or cytotoxic treatment.

Ingredients

• Allopurinol 100 mg

Direction

Adults: For mild gout, 100–200 mg daily; for moderately severe conditions, 300–600 mg daily; for severe conditions, 700–900 mg daily. Take after meals. Ensure adequate fluid intake (at least 2 litres/day). Dose should be adjusted based on serum uric acid levels.

Side effects

• Uncommon: skin rash
• nausea
• vomiting
• diarrhoea.
• Rare: severe hypersensitivity reactions including Stevens
• Johnson syndrome and toxic epidermal necrolysis (stop immediately).
• Rare: hepatotoxicity
• peripheral neuropathy.
• Uncommon: drowsiness
• headache
• taste disturbance.
• Rare: blood disorders (thrombocytopenia
• agranulocytosis).

Precautions

• Do not initiate during an acute gout attack
• wait until attack fully resolves.
• Hypersensitivity reactions can be severe (Stevens
• Johnson syndrome
• toxic epidermal necrolysis)
• stop immediately if rash develops.
• Increased risk of severe cutaneous reactions in patients carrying the HLA
• B*5801 allele (particularly in Han Chinese
• Thai
• and Korean populations) — genetic testing recommended before initiation.
• Reduce dose in renal impairment.
• Use with caution in hepatic impairment.
• Maintain adequate hydration.
• Caution with concomitant use of azathioprine or mercaptopurine (dose reduction of these agents required).
• Caution with warfarin — may enhance anticoagulant effect.
